Bacillus thuringiensis produces its insecticidal Cry protein as an inactive protoxin; activation requires the alkaline conditions of an insect's gut, so the toxin stays harmless while still inside the bacterium.

Bacillus thuringiensis (Bt) produces crystal (Cry) proteins encoded by cry genes. Inside the bacterium, this protein exists in an inactive protoxin form — it is not toxic in this state.

When an insect larva feeds on plant tissue (or Bt formulation) containing these protein crystals, the crystals are ingested and enter the insect's gut. The insect gut has a strongly alkaline pH, which solubilises and activates the protoxin, and specific gut proteases then cleave it into the active toxin fragment. This activated toxin binds to specific receptors on the insect midgut epithelial cells, creating pores that cause the cells to swell and lyse (osmotic imbalance), killing the insect.
