Protein digestion begins in the stomach, where HCl activates pepsin, which breaks down proteins into smaller polypeptides (proteoses and peptones).

The stomach's gastric glands secrete gastric juice, which contains three main components relevant to protein digestion: hydrochloric acid (HCl), the inactive proenzyme pepsinogen, and mucus.

Step 1 - Activation of pepsinogen:

HCl secreted by the oxyntic (parietal) cells maintains a highly acidic pH (around 1.5-2.5) in the stomach. This acidic environment converts the inactive pepsinogen (secreted by chief/peptic cells) into its active form, pepsin. HCl also dissolves the bits of food and kills any bacteria present.

Step 2 - Action of pepsin:

Pepsin is a proteolytic (protein-digesting) enzyme that acts on the peptide bonds of proteins, breaking down large protein molecules into smaller fragments called proteoses and peptones (partially digested polypeptides). This is only partial digestion - proteins are not fully broken down into amino acids in the stomach.

Step 3 - Role of rennin (in infants):

Infants also secrete an enzyme called rennin, which specifically acts on the milk protein casein, curdling/coagulating it into paracasein in the presence of Ca2+ ions, making it easier for pepsin to act upon.
