Capacitation is the functional maturation of sperm that occurs inside the female reproductive tract, and is essential before a sperm can fertilise an egg.

When sperm are ejaculated into the female reproductive tract during coitus, they are motile but not yet capable of fertilising the ovum. As the sperm travel through the female tract (cervix, uterus and oviduct), they undergo a series of physiological and biochemical changes collectively called capacitation.

During capacitation:

  • Glycoprotein and seminal plasma components coating the sperm's acrosomal membrane are removed/modified.
  • The plasma membrane of the sperm head becomes destabilised and more fluid, priming the acrosome.
  • The sperm's motility pattern changes (hyperactivation), giving it a more vigorous, whip-like motion needed to penetrate the egg's coverings. …
