The TCA cycle is amphibolic because it both breaks down acetyl CoA to release energy and supplies its own intermediates for biosynthesis.

Respiration is usually described as a catabolic process, since its overall purpose is to break down organic substrates and release the energy stored in their bonds. Taken purely as an energy-releasing sequence, the citric acid cycle fits this description well: acetyl CoA condenses with oxaloacetic acid to form citric acid, and a series of oxidation and decarboxylation steps completely breaks the acetyl group down, releasing CO2 and capturing energy as ATP (via GTP), NADH and FADH2.

However, the citric acid cycle does not only run in this breakdown direction. Its intermediates are also drawn upon when the cell needs to build new molecules rather than break old ones down.

  • Acetyl CoA, the point at which the cycle begins, is the same molecule the cell withdraws from the pathway when it needs to synthesise fatty acids.
  • Amino acid metabolism is similarly linked to the cycle: amino acids can be degraded to enter the cycle at various points, and by the same logic, cycle intermediates can be drawn upon in the reverse direction to support amino acid synthesis. …
