Restriction enzymes cut DNA at specific recognition sequences — the 'molecular scissors' of genetic engineering, used to isolate genes and build recombinant DNA.

Meaning. A restriction enzyme (restriction endonuclease) is an enzyme, obtained from bacteria, that recognises a specific short palindromic sequence in a DNA molecule and cuts the two strands at or near that site. Each enzyme has its own recognition sequence — for example EcoRI recognises and cuts at 5′-GAATTC-3′, leaving 'sticky ends' with unpaired overhangs. (Endonucleases cut within the DNA; exonucleases remove nucleotides from the ends.)
