rDNA technology = cut a gene with restriction enzymes, join it to a vector with ligase, put it into a host, and let the host make the desired product.

Concept. Recombinant DNA technology (genetic engineering) is a group of techniques by which a gene from one organism is inserted into the DNA of another to obtain a useful product or trait. It overcomes the limitation of ordinary breeding by transferring specific genes across species.

Tools required:

  • Restriction endonucleases — 'molecular scissors' that cut DNA at specific palindromic sequences, leaving sticky ends.
  • DNA ligase — 'molecular glue' that joins DNA fragments.
  • Cloning vectors — carriers such as plasmids or bacteriophages that replicate inside the host and carry the foreign gene (they have an ori, selectable markers and recognition sites).
  • Host organism — e.g. E. coli, yeast, that takes up and multiplies the rDNA.

Main steps:

  1. Isolation of the desired DNA/gene (and of vector DNA), followed by purification.
  2. Cutting (restriction digestion): both the gene of interest and the vector are cut with the same restriction enzyme, producing complementary sticky ends.
  3. Joining (ligation): the foreign gene is joined to the vector by DNA ligase, forming the recombinant DNA molecule.
  4. Transfer into the host (transformation): the rDNA is introduced into a competent host cell (e.g. by making cells competent with Ca²⁺, heat shock, micro-injection, or a gene gun). …
