Transcription is the copying of a DNA template strand into complementary RNA by RNA polymerase, in three steps: initiation, elongation and termination.

Definition: Transcription is the process by which the genetic information stored in a segment (gene) of DNA is copied into a complementary RNA (mRNA) molecule. Only one strand of the DNA (the template / antisense strand, 3′→5′3' \to 5') is transcribed; the other is the coding/sense strand.

Enzyme: RNA polymerase (in prokaryotes one RNA polymerase; in eukaryotes RNA Pol I, II, III).

Base-pairing rule: RNA is built with A, U, G, C. Against the DNA template — DNA A → RNA U, DNA T → RNA A, DNA G → RNA C, DNA C → RNA G (thymine is replaced by uracil in RNA).

Steps:

  1. Initiation: RNA polymerase binds to the promoter on DNA; the double helix unwinds/opens up.
