Denaturation = loss of a protein's native 2°/3° structure (and hence function) with the 1° structure intact. Nucleic-acid bases = purines A, G and pyrimidines C, T (DNA), U (RNA).

(i) Denaturation of proteins. A protein in its biologically active form has a specific native conformation held by hydrogen bonds, disulphide links and other weak interactions (its secondary and tertiary structure). When the protein is subjected to heat, strong acid or base, heavy-metal salts, alcohol, etc., these weak forces are disrupted and the native shape is lost. This process is called denaturation.

  • The primary structure (the sequence joined by peptide bonds) remains unchanged.
  • The secondary and tertiary structures are destroyed, so the protein loses its biological activity.

Examples: coagulation of egg white (albumin) on boiling, and curdling of milk.

(ii) Bases present in nucleic acids. Nucleic acids contain two kinds of nitrogenous bases:

  • Purines (double-ring):
    • Adenine (A) — a purine with an −NH2-NH_2 group at position 6.
    • Guanine (G) — a purine with a keto (=O=O) group and an −NH2-NH_2 group.
  • Pyrimidines (single-ring):
    • Cytosine (C) — pyrimidine with an −NH2-NH_2 group (present in both DNA and RNA).
    • Thymine (T) — 5-methyl pyrimidine with two keto groups (present in DNA only).
