The promoter (a DNA sequence at the 5′ end of a gene) (1) is the binding site for RNA polymerase and (2) defines the template strand and the start/direction of transcription.

Concept

A transcription unit in DNA has three parts: a promoter, the structural gene, and a terminator. The promoter is located towards the 5′ end (upstream) of the coding sequence of the structural gene.

Two functions of the promoter

  1. It provides the recognition and binding site for RNA polymerase — RNA polymerase attaches at the promoter to initiate transcription.
  2. It defines the template strand and the direction of transcription — because the promoter lies at one end, it fixes which of the two DNA strands acts as the template and where transcription starts (giving transcription its polarity).
