Bt insecticidal protein is an inactive crystal that the insect's alkaline gut activates; it then punctures the gut cells and kills the pest.

The soil bacterium Bacillus thuringiensis produces the insecticidal protein as inactive crystalline protoxin (Cry protein). Mechanism of action:

  1. A susceptible insect (e.g. bollworm, corn borer) ingests the crystals along with plant tissue.
  2. In the insect's midgut the pH is highly alkaline, which dissolves the crystals and, aided by gut proteases, converts the protoxin into the active toxin.
  3. The active Cry toxin binds to specific receptors on the surface of the midgut epithelial cells.
  4. It creates pores in the cell membranes; the cells swell, take in water, lyse and the gut is destroyed. The insect stops feeding and dies.

Because activation needs an alkaline gut, the toxin is harmless to humans and animals (whose stomachs are acidic). Genes for these proteins (cryIAc, cryIIAb, cryIAb) are introduced into crops such as Bt cotton and Bt corn for pest resistance.
