The zona pellucida is a transparent, glycoprotein-rich membrane that surrounds the mammalian ovum. While it plays several roles during fertilization—acting as a species-specific barrier and providing binding sites for sperm—its most critical function emerges immediately after the first sperm penetrates: preventing polyspermy, the potentially lethal condition where multiple sperm fertilize a single egg.

When a sperm successfully binds to and penetrates the zona pellucida, it fuses with the plasma membrane of the ovum. This fusion is the trigger. The moment sperm-egg fusion occurs, the egg responds with what is called the cortical reaction. Cortical granules, which are membrane-bound vesicles lying just beneath the egg's plasma membrane, immediately release their contents into the space between the plasma membrane and the zona pellucida. This release happens within seconds of the first sperm's entry.

The enzymes released from these cortical granules—particularly proteases and peroxidases—modify the zona pellucida in two crucial ways. First, they alter the glycoprotein structure of the zona, specifically changing the sperm-binding receptors (like ZP3) so that additional sperm can no longer recognize or attach to them. Second, these enzymes cause the zona pellucida to harden, a process sometimes called the zona reaction.
