To initiate translation, the messenger RNA (mRNA) first binds to the smaller ribosomal subunit.

Translation is the fundamental biological process by which the genetic information encoded in messenger RNA (mRNA) is decoded to synthesize proteins. This intricate process occurs on ribosomes, which are complex molecular machines found in the cytoplasm of all living cells. Understanding the initial steps of translation is crucial, as it sets the stage for accurate protein synthesis.

Ribosomes themselves are composed of ribosomal RNA (rRNA) and numerous proteins, existing in two main parts: a large ribosomal subunit and a small ribosomal subunit. In their inactive state, these two subunits remain separate. The initiation of translation is a highly regulated process that brings together the mRNA template, the initiator transfer RNA (tRNA) carrying the first amino acid, and the ribosomal subunits.

The critical first step in initiating translation involves the mRNA molecule. Rather than binding to the entire ribosome directly or to the larger subunit, the mRNA specifically seeks out and binds to the smaller ribosomal subunit.

  • In prokaryotic cells (like bacteria), the smaller ribosomal subunit identifies and binds to a specific sequence on the mRNA, known as the Shine-Dalgarno sequence, which is located just upstream of the start codon (AUG).
  • In eukaryotic cells (like plants and animals), the smaller ribosomal subunit, along with several initiation factors and the initiator tRNA (carrying methionine), first binds to the 5' cap structure of the mRNA. It then scans along the mRNA until it locates the start codon (AUG). …
