Glycolysis is the cytoplasmic, oxygen-independent, ten-step scheme (EMP pathway) that converts one glucose into two molecules of pyruvic acid, with a net gain of 2 ATP and 2 NADH.

Glycolysis (from the Greek glycos = sugar, lysis = splitting) was given by Embden, Meyerhof and Parnas (the EMP pathway). It occurs in the cytoplasm of all living organisms and does not require oxygen.

Schematic representation

Glucose
   | (ATP used)
Glucose-6-phosphate
   |
Fructose-6-phosphate
   | (ATP used)
Fructose-1,6-bisphosphate
   | (splits into two 3-carbon sugars)
Dihydroxyacetone phosphate  <-->  Glyceraldehyde-3-phosphate (PGAL)
                                     | (NAD+ -> NADH + H+)
                              1,3-bisphosphoglyceric acid (BPGA)
                                     | (ATP formed)
                              3-phosphoglyceric acid (PGA)
                                     |
                              2-phosphoglyceric acid
                                     | (water removed)
                              Phosphoenolpyruvate (PEP)
                                     | (ATP formed)
                              Pyruvic acid

Key points

  • Two ATP are consumed in the early steps (forming glucose-6-phosphate and fructose-1,6-bisphosphate).
  • The 6-carbon fructose-1,6-bisphosphate splits into two 3-carbon molecules; the dihydroxyacetone phosphate is interconvertible with glyceraldehyde-3-phosphate, so effectively two PGAL proceed.
  • Two NADH + H+ are produced when PGAL is oxidised.
  • Four ATP are formed by substrate-level phosphorylation (2 at the BPGA to PGA step, 2 at the PEP to pyruvate step).
  • Net gain = 4 ATP formed − 2 ATP used = 2 ATP, plus 2 NADH + H+, and two molecules of pyruvic acid per glucose.
✓Final answer

Glucose → glucose-6-phosphate → fructose-6-phosphate → fructose-1,6-bisphosphate → (split) DHAP + PGAL → BPGA → PGA → 2-phosphoglycerate → PEP → pyruvic acid (two molecules); net yield per glucose = 2 ATP and 2 NADH + H+.
