Glucose is phosphorylated by hexokinase (using ATP) to glucose-6-phosphate, which is isomerized to fructose-6-phosphate, then phosphorylated again by phosphofructokinase (using a second ATP) to fructose-1,6-bisphosphate. This 6-carbon compound is split into two 3-carbon molecules: dihydroxyacetone phosphate (DHAP) and glyceraldehyde-3-phosphate (G3P), which are interconvertible. Each G3P is oxidized (NAD+ to NADH) and phosphorylated to 1,3-bisphosphoglycerate, which is converted to 3-phosphoglycerate generating one ATP (substrate-level phosphorylation); this proceeds to 2-phosphoglycerate, then dehydrated to phosphoenolpyruvate (PEP), which transfers its phosphate to ADP forming pyruvate and another ATP. Since two G3P molecules are processed per glucose, the net yield of glycolysis is 2 ATP, 2 NADH and 2 pyruvate molecules per glucose, occurring entirely in the cytoplasm without requiring oxygen.
