Mature insulin is a two-chain hormone (A chain + B chain, linked by disulfide bonds); its precursor, proinsulin, has these two chains joined by an extra connecting 'C peptide' that is cut out during maturation. Biotechnologically, the A and B chains are made separately in engineered E. coli and then joined to form functional insulin.

Structure of proinsulin. In the body, insulin is first synthesised as a single-chain precursor called proinsulin, made of three segments joined end to end: the A chain, the connecting C peptide in the middle, and the B chain. The C peptide holds the A and B chains in the correct three-dimensional arrangement so that the disulfide bonds between the A and B chains form correctly. Once these bonds are in place, an enzyme cleaves out the C peptide, leaving mature, active insulin — the A chain and B chain now held together only by disulfide bonds (the C peptide is absent from mature insulin).

Production of mature insulin through rDNA technology. Before genetic engineering, insulin was extracted from the pancreas of slaughtered cattle and pigs; this animal insulin sometimes caused allergic reactions. Eli Lilly (an American company) then prepared genetically engineered (recombinant) insulin, marketed as Humulin, as follows:
