Insulin is made as proinsulin and needs the C-peptide removed before it becomes biologically active.

In the human body, insulin is synthesised as a pro-hormone which must be processed before it becomes a mature and functional hormone. Proinsulin contains an extra stretch of amino acids called the C-peptide, in addition to the A-chain and B-chain that make up mature insulin. During processing in the pancreatic β-cells, the C-peptide is enzymatically excised, and the remaining A-chain and B-chain become linked together by disulphide bonds, forming the mature, biologically active insulin molecule.
