The endoplasmic reticulum is a network of membranous tubules/cisternae that synthesises and transports proteins (rough ER) and lipids (smooth ER), besides contributing to detoxification and membrane biogenesis.

The endoplasmic reticulum (ER) is present in the cytoplasm of eukaryotic cells and occurs as two types — rough ER (RER, studded with ribosomes) and smooth ER (SER, lacking ribosomes). Its main functions include:

  1. Protein synthesis and processing: The ribosomes attached to RER actively synthesise proteins, which are then processed (folding, initial glycosylation) and transported through the ER lumen to their destinations, e.g., to the Golgi apparatus for further processing/packaging.
  2. Lipid synthesis: SER is the major site of synthesis of lipids, including cell membrane phospholipids, steroids/steroid hormones and cholesterol.
  3. Forms cell membrane components: The ER manufactures almost all the lipids and some proteins required for building new cell membranes, thereby playing a central role in membrane biogenesis (e.g., contributing membrane material to the Golgi apparatus, and, indirectly, the plasma membrane). …
