RER is ribosome-studded and specialises in protein synthesis, while SER is ribosome-free and specialises in lipid synthesis and detoxification.

The endoplasmic reticulum (ER) is a network of membranous tubules and sacs (cisternae) within the cytoplasm, of which there are two functionally and structurally distinct forms:

  1. Presence of ribosomes: Rough endoplasmic reticulum (RER) has numerous ribosomes attached to its cytoplasmic (outer) surface, giving it a granular, 'rough' appearance under the electron microscope. Smooth endoplasmic reticulum (SER) has no ribosomes attached to its surface, giving it a smooth appearance.

  2. Shape: RER is typically organised as flattened, sac-like cisternae. SER is more often organised as a branching network of tubules and small vesicles.
