Root nodules form when nitrogen-fixing Rhizobium bacteria infect a legume's root hairs, travel inward via an infection thread, and multiply in the cortex, causing the cortical cells to proliferate into a nodule where the bacteria fix atmospheric nitrogen symbiotically with the plant.

Nodule formation is a classic example of symbiosis, seen typically in the roots of leguminous (family Fabaceae) plants such as pea, gram, and beans.

Steps in nodule formation:

  1. Free-living Rhizobium bacteria present in the soil are attracted to and colonise the root hair region of a legume root, in response to chemical signals (flavonoids) released by the root.
  2. The bacteria attach to and infect a root hair, which curls around the bacteria; the bacteria enter through the root hair by producing an infection thread, a tubular ingrowth of the root hair cell membrane.
  3. The infection thread carries the bacteria into the cells of the root cortex.
  4. Once inside, the bacteria multiply rapidly and stimulate the cortical cells to divide and enlarge, producing a swelling — the root nodule.
