Root nodule formation is a step-wise infection process — chemical recognition, root-hair curling, infection thread formation, cortical cell division, and bacteroid differentiation — that ends with the bacteria fixing atmospheric nitrogen for the plant.

Certain soil bacteria, mainly of the genus Rhizobium, form a mutually beneficial (symbiotic) relationship with the roots of leguminous plants, resulting in the formation of root nodules where nitrogen fixation takes place. The steps are:

  1. Recognition — free-living Rhizobium bacteria in the soil are chemically attracted to the root hairs of a compatible host legume by substances (e.g., flavonoids) exuded by the roots; the bacteria in turn produce Nod factors recognised by the root.
  2. Root hair curling — upon recognition, the root hair curls, trapping the attached bacteria.
  3. Infection thread formation — the plant cell wall and membrane invaginate to form a tubular "infection thread" through which the bacteria travel inward, from the root hair into the root cortex.
  4. Cortical cell division — the presence of the invading bacteria stimulates the cells of the root cortex to divide repeatedly, producing the swelling that becomes the visible root nodule. …
