Muscle contraction occurs by the sliding filament theory: calcium-triggered cross-bridge cycling between myosin and actin pulls the thin filaments inward, shortening the sarcomere.

Each skeletal muscle fibre contains many myofibrils, which are made of repeating contractile units called sarcomeres, containing overlapping thick filaments (myosin) and thin filaments (actin, with the regulatory proteins troponin and tropomyosin). The steps of contraction are:

  1. A nerve impulse arrives at the neuromuscular junction, releasing the neurotransmitter acetylcholine, which generates an action potential in the muscle fibre membrane (sarcolemma).
  2. This action potential travels into the muscle fibre via T-tubules and triggers the sarcoplasmic reticulum to release stored calcium ions (Ca2+) into the sarcoplasm.
  3. Ca2+ binds to troponin on the thin filament, causing tropomyosin to shift and expose the myosin-binding sites on actin.
  4. Myosin heads (cross-bridges), energised by ATP hydrolysis, attach to these exposed sites on actin and pull the thin filaments toward the centre of the sarcomere (the 'power stroke').
  5. ATP binding to myosin causes the cross-bridge to detach, and its hydrolysis re-cocks the myosin head for another cycle - this cross-bridge cycling repeats rapidly as long as Ca2+ and ATP are available. …
