Enzymes are the protein catalysts themselves; co-enzymes are separate, non-protein 'helper' organic molecules that must associate with certain enzymes for them to function.

FeatureEnzymeCo-enzyme
Chemical natureA protein (globular, has primary/secondary/tertiary structure)A non-protein organic molecule, often derived from a vitamin (e.g. NAD⁺, FAD, coenzyme A)
RoleActs as the actual biocatalyst — provides the active site that binds the substrate and lowers the activation energy of the reactionBinds to the enzyme (usually loosely/reversibly) as a cofactor, assisting catalysis, often by accepting/donating specific chemical groups (e.g. hydrogen atoms, acyl groups) during the reaction
DependenceSome enzymes work alone; others (called 'holoenzymes' when complete) absolutely require a co-enzyme to be catalytically activeHas no catalytic activity by itself — only functions when bound to its specific enzyme (apoenzyme)
Size/complexityLarge, complex, folded macromoleculeRelatively small organic molecule
