Cloning vectors are self-replicating DNA carriers (plasmids/phages) used to introduce and multiply a foreign gene inside a host cell.

In recombinant DNA technology, once a gene of interest has been cut out using restriction enzymes, it needs to be carried into a host cell and made to replicate along with the host - this carrier DNA molecule is called a cloning vector. Commonly used cloning vectors include bacterial plasmids (small, circular, extrachromosomal, self-replicating DNA molecules, e.g. pBR322) and bacteriophages (viruses that infect bacteria). For a DNA molecule to function efficiently as a cloning vector, it must have: an origin of replication (ori) that allows autonomous replication inside the host and controls copy number; a selectable marker (such as genes for antibiotic resistance, e.g. ampicillin or tetracycline resistance) that helps identify and eliminate untransformed …
