During DNA replication, the leading strand is made continuously toward the replication fork, while the lagging strand is made discontinuously (in Okazaki fragments) away from the fork and must later be sealed by DNA ligase.

DNA replication is semi-conservative and occurs at a Y-shaped replication fork, where the two parental strands are unwound and each is used as a template. Because DNA polymerase can only synthesise a new strand in the 5'→ 3' direction (adding nucleotides only to a free 3'-OH end), and the two parental template strands run antiparallel to each other, the two new strands must be synthesised differently at the same fork:
