DNA replication is semiconservative: the two strands separate, each serves as a template, and DNA polymerase synthesises a complementary strand 5'->3' (leading strand continuous, lagging strand in Okazaki fragments), giving two DNAs each with one old and one new strand.

DNA replication is the process by which a DNA molecule makes an exact copy of itself before cell division. Meselson and Stahl (1958) proved it is semiconservative - each daughter molecule retains one parental (old) strand and one newly synthesised strand.

Steps:

  1. Origin and unwinding: Replication begins at a specific site called the origin of replication. The enzyme helicase unwinds and separates the two strands, forming a Y-shaped replication fork; single-strand binding proteins keep the strands apart and topoisomerase (gyrase) relieves the coiling tension ahead of the fork.

  2. Template and priming: Each separated strand acts as a template. The enzyme DNA polymerase cannot start on its own, so a short RNA primer laid down by primase provides a free 3'-OH end.

  3. Synthesis of new strands: DNA polymerase III adds deoxyribonucleotides to the 3' end, so a new strand always grows in the 5'->3' direction, following complementary base pairing (A with T, G with C). Because the two template strands are antiparallel:

    • The leading strand (template 3'->5') is synthesised continuously towards the fork.
    • The lagging strand (template 5'->3') is synthesised discontinuously, away from the fork, as short pieces called Okazaki fragments. …
