Denaturation disrupts the hydrogen bonds and other weak interactions holding a protein's 3-D shape, unfolding it and destroying its biological function, without breaking the peptide bonds of its primary structure.

A protein's biological activity depends critically on its native three-dimensional conformation (its secondary structure - alpha-helix/beta-pleated sheet - and tertiary structure, the overall folded shape), which is held together by hydrogen bonds and other weak interactions between amino acid side chains.

When a native protein is subjected to a physical change such as heating, or a chemical change such as a change in pH (addition of acid/base) or treatment with certain reagents, these weak stabilising interactions (hydrogen bonds, etc.) are disrupted. As a result, the globular protein unfolds and its specific 3-D shape is lost - the polypeptide chain typically becomes a random, extended (fibrous-like) structure. This process is called denaturation.
