  • (b) Primers - annealing: two short, chemically synthesised single-stranded oligonucleotides complementary to the two ends of the target region. At a lower temperature they base-pair (anneal) with the template strands and provide a free 3'-OH group, which DNA polymerase needs to start synthesis; they also define exactly which segment is copied.
  • (c) Thermus aquaticus - extension: this thermophilic bacterium is the source of Taq DNA polymerase, a thermostable enzyme that remains active at the high denaturation temperature.
