A restriction enzyme's staggered cut across a palindromic DNA sequence generates single-stranded 'sticky' overhangs that can base-pair with any DNA cut by the same enzyme, and DNA ligase then permanently seals the join.

Formation of sticky ends: Restriction endonucleases (e.g. EcoRI) recognise and bind to a specific, short palindromic nucleotide sequence in double-stranded DNA (a sequence that reads the same on both strands in the 5'→3' direction). Instead of cutting straight across both strands at the exact centre of this sequence, the enzyme makes a staggered cut, cleaving each strand between the same two bases on their respective strands, but offset from the centre. This staggered cut leaves short, single-stranded overhanging ends at each cut fragment — these are called sticky ends, because their exposed, unpaired bases are complementary and can readily base-pair (hydrogen-bond) with any other DNA fragment that has been cut by the same restriction enzyme (since it will generate a matching complementary overhang).
