Crossing over — the exchange of genetic material between non-sister chromatids of homologous chromosomes — occurs during pachytene, the third sub-stage of meiotic prophase I.

Prophase I of meiosis is long and is divided into five sub-stages: Leptotene → Zygotene → Pachytene → Diplotene → Diakinesis.

  • In leptotene, chromosomes begin to condense.
  • In zygotene, homologous chromosomes pair up (synapsis), forming bivalents/tetrads, and the synaptonemal complex forms.
  • In pachytene, the bivalent chromosomes show clearly as tetrads (four chromatids). Crossing over takes place here: non-sister chromatids of the paired homologous chromosomes exchange segments at points called chiasmata, mediated by recombination nodules containing the enzymes needed for breakage and reunion of DNA. …
