The origin of replication (ori) is an essential feature of any cloning vector, as it is the sequence that enables the linked (foreign) DNA to be copied inside the host cell.

A cloning vector is a DNA molecule (such as a plasmid or bacteriophage) used to carry a foreign DNA fragment into a host cell and replicate it there. One key feature required in a cloning vector is:

Origin of replication (ori): This is a specific sequence in the vector DNA from where DNA replication begins. Any piece of source DNA that gets linked (ligated) to this vector sequence can be made to replicate within the host cell, because replication is controlled by sequences at the ori. The origin also controls the copy number of the linked DNA - vectors with a high copy-number origin will produce many copies of the recombinant DNA per host cell, which is useful for obtaining large amounts of the cloned DNA.
