A son with red-green colour blindness cannot inherit the trait from his father because the gene for this condition is located on the X chromosome, and a father passes his Y chromosome to his son, not his X chromosome.

Red-green colour blindness is a classic example of an X-linked recessive disorder. To understand why a son cannot inherit it from his father, we need to look at how sex chromosomes are passed from one generation to the next. In humans, females have two X chromosomes (XX), while males have one X and one Y chromosome (XY). A son always receives his Y chromosome from his father and his X chromosome from his mother. This is the fundamental biological rule that governs the inheritance pattern.

The gene responsible for red-green colour vision is located on the X chromosome. Because it is recessive, a female needs two copies of the defective allele (one on each of her X chromosomes) to be colour-blind. A male, however, has only one X chromosome, so a single defective allele on that X chromosome will cause colour blindness. This is why the condition is far more common in males than in females.

Now, consider the father in the question. He is colour-blind, meaning his single X chromosome carries the defective allele. When he has a son, he contributes his Y chromosome — not his X chromosome. The son’s X chromosome comes from the mother. Therefore, the father cannot pass the colour-blindness allele to his son. The son’s colour blindness must have come from his mother, who is either a carrier (heterozygous) or herself colour-blind. …
