Denaturation happens when physical or chemical agents (heat, acid/base, heavy metals, etc.) disrupt the weak interactions holding a protein's 3-D folded shape, unfolding it into a random coil and destroying its biological function, without breaking the peptide bonds of the primary structure.

Every protein has a specific 3-dimensional shape (its native/biologically active conformation), maintained by weak, non-covalent interactions such as hydrogen bonds, and by disulfide bridges, which hold its secondary structure (alpha-helix/beta-sheet) and tertiary structure (overall folding) in place.

When a protein in its native form is subjected to a physical change (like heating) or a chemical change (like a change in pH, addition of certain salts, or treatment with acids/bases), these weak interactions holding the secondary and tertiary structure are disrupted. The globules of the protein unfold and the helix uncoils - this is called denaturation.

During denaturation, ONLY the secondary and tertiary structures are destroyed (the folded shape is lost); the primary structure (the actual sequence of amino acids joined by peptide bonds) remains unaffected/intact.
