The endoplasmic reticulum (ER) is a network of membranous tubules and cisternae in the cytoplasm, occurring in two forms:

Rough Endoplasmic Reticulum (RER):

  • Has numerous ribosomes attached to the outer surface of its membrane, giving it a 'rough' or granular appearance.
  • Is the principal site of protein synthesis; proteins synthesised on the attached ribosomes are threaded into the ER lumen for folding, modification, and eventual packaging/secretion.
  • Is more abundant in cells actively engaged in protein secretion, e.g., pancreatic cells.
  • Often continuous with the outer nuclear membrane on one side and the Golgi apparatus on the other.
