Bacterial conjugation is a form of genetic recombination in which DNA is transferred directly from a donor cell to a recipient cell through physical contact, typically via a plasmid such as the F (fertility) factor.

Discovery:

Conjugation was first demonstrated by Lederberg and Tatum in Escherichia coli.

Mechanism:

  1. A donor cell possessing the F (fertility) plasmid is called F+; a recipient cell lacking it is F-.
  2. The F+ cell develops a hair-like appendage called the sex pilus (F-pilus), which makes contact with an F- recipient cell and draws the two cells together, forming a conjugation bridge/tube.
  3. At the origin of transfer (oriT) on the F plasmid, one strand of the plasmid DNA is nicked and begins to unwind.
  4. The single strand of plasmid DNA is transferred through the conjugation tube into the recipient cell, while a new complementary strand is synthesised in the donor cell to restore its double-stranded plasmid.
  5. Inside the recipient cell, a complementary strand is also synthesised, converting the recipient into an F+ cell as well. …
