EcoRI is a sequence-specific restriction enzyme that cuts double-stranded DNA at GAATTC sites, leaving single-stranded sticky ends used to join DNA fragments in genetic engineering.

What it is: EcoRI is a restriction endonuclease, first isolated from the bacterium Escherichia coli (hence the name: E.coli, R = restriction, I = first identified). It belongs to the class of restriction enzymes that recognize a specific, short, palindromic DNA sequence.

How it functions: EcoRI recognizes the palindromic sequence:

5'-G A A T T C-3'

3'-C T T A A G-5'

It cuts each strand between the G and the adjacent A (i.e., between G and A on both strands), producing fragments with short, single-stranded, complementary overhangs called 'sticky ends':

5'-G AATTC-3'
