Root nodules form through a step-by-step signalling and infection process between Rhizobium bacteria and legume root hairs, culminating in a nitrogen-fixing symbiotic organ.

Root nodules are specialized structures formed on the roots of leguminous plants as a result of symbiosis with nitrogen-fixing bacteria of the genus Rhizobium.

Steps in root nodule formation:

  1. Chemical signalling/attraction: The legume root secretes chemical compounds called flavonoids into the surrounding soil, which attract free-living Rhizobium bacteria present in the soil towards the root.

  2. Attachment to root hairs: The Rhizobium bacteria attach themselves to the root hairs of the host plant.

  3. Root hair curling: In response to signalling molecules (Nod factors) secreted by the bacteria, the root hair curls, and the bacteria become trapped within the curl.

  4. Infection thread formation: The bacteria induce the formation of an infection thread — a tubular ingrowth of the root hair's plasma membrane — through which the bacteria travel/invade into the root cortex cells.

  5. Cortical cell division and nodule development: The presence of the bacteria stimulates the cortical cells of the root to divide rapidly, forming a swelling that develops into the visible root nodule.

  6. Bacteroid formation: Inside the nodule cells, the bacteria differentiate into a modified, enlarged, often branched non-reproductive form called bacteroids, enclosed within a plant-derived peribacteroid membrane forming structures called symbiosomes.
