For growing a plant explant in vitro (tissue culture), it must be provided with a nutrient medium that supplies everything the isolated tissue needs to survive, divide and differentiate, since it cannot obtain these from the whole plant or soil. A commonly used medium is the Murashige and Skoog (MS) medium, which generally contains the following components:

  • Inorganic nutrients (mineral salts): Both macronutrients (required in larger amounts, e.g., nitrogen, phosphorus, potassium, calcium, magnesium, sulphur) and micronutrients (required in trace amounts, e.g., iron, manganese, zinc, copper, boron, molybdenum), which are essential for normal growth and metabolism, just as they are for whole plants.
  • Carbon and energy source: Usually sucrose, since the explant, grown in a closed vessel with limited or no light and often not fully photosynthetically active, cannot make its own food and depends on an external supply of sugar for energy.
  • Vitamins: Such as thiamine, nicotinic acid, pyridoxine and others, which act as co-factors in various metabolic reactions and support healthy growth of the culture.
  • Amino acids and nitrogen supplements: Sometimes added (e.g., glycine) as an additional organic nitrogen source to promote growth.
  • Plant growth regulators (hormones): Auxins (e.g., 2,4-D) and cytokinins, added in specific concentrations and ratios to induce cell division, callus formation, and to direct differentiation of the callus into roots, shoots or somatic embryos as desired. …
