The endodermis is a single, continuous layer of cells forming the innermost boundary of the root cortex, lying immediately outside the vascular tissue (pericycle and stele). It plays a critical, highly specific role in controlling mineral absorption.

Structural basis — the Casparian strip:

  • The radial and transverse walls of endodermal cells (but notably not their tangential walls, i.e., the walls parallel to the root surface) are impregnated with a band of suberin, a waxy, hydrophobic substance (and often lignin as well), forming a structure called the Casparian strip.
  • This strip is completely impermeable to water and dissolved mineral ions.

Functional consequence:

  • Up to the endodermis, water and minerals absorbed by the root can move through the cortex via two parallel pathways: the apoplast (through the continuum of cell walls and intercellular spaces, without crossing any membrane) and the symplast (through the interconnected cytoplasm of cells via plasmodesmata, requiring passage across at least one membrane to first enter).
  • On reaching the endodermis, the Casparian strip physically blocks the apoplastic pathway — water and minerals travelling through the cell walls cannot pass through this waxy barrier.
  • As a result, at the endodermis, all water and minerals must enter the symplast — that is, they must cross the selectively permeable plasma membrane of the endodermal cells and pass through their cytoplasm — before being able to re-emerge (via the inner tangential wall or plasmodesmata leading to the pericycle) and finally enter the xylem.
