Actin is the smaller, thin-filament protein (I-band); myosin is the larger, thick-filament protein (A-band) with ATPase-bearing heads that form cross-bridges and pull actin during contraction.

Skeletal muscle contraction depends on two major contractile proteins - actin and myosin - arranged as myofilaments in the myofibrils. Their differences are:

Actin (thin filament):

  • Forms the thin filaments of the myofibril.
  • It is a smaller and lighter protein; globular G-actin units polymerise to form filamentous F-actin, two of which twist to form the thin filament, associated with tropomyosin and troponin.
  • Located in the I-band (and extends into the A-band).
  • Acts as the 'track' along which myosin moves.

Myosin (thick filament):

  • Forms the thick filaments of the myofibril. …
