Human insulin genes (A and B chains) are cloned in E. coli; the chains are produced, purified and joined by disulphide bonds to give recombinant insulin.

Human insulin consists of two short polypeptide chains, A (21 amino acids) and B (30 amino acids), joined by disulphide bridges. In the body it is made as a single pro-insulin with an extra C-peptide that is removed during maturation. To produce insulin by recombinant DNA (rDNA) technology:
