Bacterial transformation is the process by which bacteria take up foreign DNA from their surroundings. In nature, this happens rarely and only in certain bacterial species under specific conditions. In the laboratory, scientists have developed methods to make bacteria "competent" — that is, capable of taking up plasmid DNA — so they can introduce recombinant DNA into bacterial cells for cloning and gene expression.
The heat shock method is a critical step in this artificial transformation protocol. Before heat shock, bacterial cells are treated with calcium chloride at low temperature (around 4°C). This treatment makes the cell membrane more permeable and prepares the cells to accept DNA. The plasmid DNA is then mixed with these competent cells while they remain cold.
The actual heat shock involves a sudden, brief exposure to elevated temperature — typically 42°C for about 90 seconds — followed by an immediate return to ice. This thermal jolt is not about binding DNA to the surface or activating transport proteins. Instead, the rapid temperature change causes a physical disruption in the bacterial cell wall and membrane structure. The cell wall momentarily develops tiny openings or pores.
These transient pores are the key. During the brief window when they exist, the plasmid DNA molecules that have been sitting on the cell surface can slip through into the cytoplasm. The pores close quickly once the cells are returned to cold conditions, trapping the DNA inside.
