The C-peptide connects the A and B chains in proinsulin and is removed during processing to yield mature, active insulin.

Insulin, as synthesised in the body, initially contains an extra stretch of amino acids called the C-peptide, which is not present in the mature/active form of insulin. Proinsulin (containing the A chain, C-peptide, and B chain) needs to be processed, during which the C-peptide is enzymatically removed, allowing the A and B chains — held together by disulfide bonds — to fold correctly and form the biologically active, mature insulin hormone.
