Concept

By 1950 it was known that genes are on chromosomes, which contain both DNA and protein. Alfred Hershey and Martha Chase (1952) designed an experiment using bacteriophages to decide which of the two — DNA or protein — actually carries the genetic information into the next generation.

The experiment — step by step

  1. A bacteriophage consists of a DNA core enclosed in a protein coat. DNA contains phosphorus (P) but no sulphur; protein contains sulphur (S) but no phosphorus — this difference was the key.
  2. Phages were grown in two separate cultures on radioactive media:
    • one with radioactive phosphorus (32P^{32}P) → phages with radioactively-labelled DNA;
    • one with radioactive sulphur (35S^{35}S) → phages with radioactively-labelled protein coat.
  3. These labelled phages were allowed to infect unlabelled E. coli bacteria.
  4. Soon after infection, the phage coats were detached from the bacteria by agitating in a blender (blending), and the mixture was centrifuged to separate the heavier bacteria (pellet) from the lighter viral coats (supernatant).
