As electrons pass through Complexes I, III and IV of the electron transport system, the energy released is used to actively pump protons (H+) from the mitochondrial matrix into the intermembrane space, building up a much higher proton concentration outside the matrix than within it -- the proton-motive force. According to the chemiosmotic hypothesis, protons can re-enter the matrix only through the channel provided by ATP synthase; as they flow back down this concentration gradient, the energy released drives ATP synthase's rotary catalytic mechanism, phosphorylating ADP to ATP using inorganic phosphate.
