Chromosomal DNA = large, essential, single-copy main genome; Plasmid DNA = small, extra, self-replicating, multi-copy, used as a vector.

Differences between plasmid DNA and chromosomal DNA:

  1. Size: Chromosomal DNA is very large (contains thousands of genes). Plasmid DNA is small (a few to a few tens of genes).
  2. Essentiality: Chromosomal DNA carries genes essential for the cell's survival and normal functioning. Plasmid DNA carries additional, non-essential genes (e.g. antibiotic resistance) that give the cell extra abilities but are not needed for basic survival.
  3. Replication: Chromosomal DNA replicates once per cell division, in step with the cell cycle. Plasmid DNA has its own origin of replication and replicates independently (autonomously), often producing many copies.
