Eli Lilly's Humulin was made by producing the insulin A and B chains separately in genetically engineered E. coli, then chemically joining them via disulphide bonds to form functional human insulin.

Insulin used to treat diabetes was earlier extracted from the pancreas of slaughtered cattle and pigs — but this animal insulin, though chemically similar, differs slightly from human insulin and could cause allergic reactions in some patients.

In 1983, the American company Eli Lilly prepared biosynthetic (recombinant) human insulin using genetically engineered E. coli. Human insulin consists of two short polypeptide chains — chain A and chain B — that are linked together by disulphide bridges.

The method followed:

  1. Two separate DNA sequences, corresponding to the A chain and the B chain of human insulin, were prepared and inserted into two separate plasmids, which were introduced into (and expressed in) two different E. coli cultures.
