Eli Lilly's 1983 method bypassed the C-peptide of pro-insulin altogether by making the A and B chains of human insulin separately in two different E. coli cultures and then chemically joining them via disulphide bonds — the first genetically engineered/recombinant human insulin, marketed as Humulin.

Insulin used to treat diabetes was earlier extracted from the pancreas of slaughtered cattle and pigs, but this animal insulin, though chemically similar to human insulin, sometimes caused allergic reactions since it differs from human insulin at one or more amino acid positions. Mature, functional human insulin consists of two short polypeptide chains, chain A and chain B, that are linked together by disulphide bridges. In the body, insulin is synthesised as a pro-hormone (pro-insulin) that contains an extra connecting C-peptide, which is enzymatically removed during maturation to yield the active hormone.
