Eli Lilly's genetically engineered insulin ('Humulin') was made by producing the insulin A and B chains separately in two different strains of E. coli using recombinant DNA technology, then chemically combining the two chains via disulfide bonds to form active human insulin — eliminating the need to extract insulin from animal pancreases.

Before recombinant insulin, diabetics were treated with insulin extracted from the pancreas of slaughtered cattle and pigs, which sometimes caused allergic reactions since it differed slightly from human insulin.

Human insulin consists of two short polypeptide chains, chain A and chain B, linked together by disulfide bonds. The American company Eli Lilly used the following approach to produce genetically engineered human insulin:

  1. Two separate DNA sequences corresponding to the A and B chains of human insulin were chemically synthesised.
  2. These DNA sequences were introduced separately into plasmid vectors and inserted into two different strains of the bacterium Escherichia coli, so that each strain produced only one of the two chains.
