Recombinant DNA can be introduced directly into a host cell (bypassing a biological vector) by micro-injection or by the gene gun (biolistic) method.

Besides using biological vectors (plasmids, bacteriophages, Agrobacterium) to carry recombinant DNA into a host, there are direct, physical/chemical methods of gene transfer:

  1. Micro-injection: Recombinant DNA is directly injected into the nucleus of an animal cell using a very fine glass micro-needle/micropipette under a microscope. This method is widely used to create transgenic animals.
  2. Gene gun / biolistics (particle bombardment): DNA is coated onto the surface of tiny gold or tungsten microparticles/pellets, which are then physically shot (bombarded at high velocity) into the target plant cells or tissue using a gene gun. This method is especially useful for plant cells, which have a tough cell wall that other methods struggle to cross. …
