In the preparatory (endergonic) phase of glycolysis, ATP is consumed to phosphorylate the sugar at two points: hexokinase phosphorylates glucose to glucose-6-phosphate at step 1, and phosphofructokinase phosphorylates fructose-6-phosphate to fructose-1,6-bisphosphate at step 3.
Step 2. In the pay-off (oxidative) phase, ATP is generated by transferring a phosphate group directly from a phosphorylated substrate to ADP -- this substrate-level phosphorylation (dephosphorylation of the substrate) occurs twice: phosphoglycerate kinase converts 1,3-bisphosphoglycerate to 3-phosphoglycerate at step 7, generating ATP, and pyruvate kinase converts phosphoenolpyruvate to pyruvate at step 10, generating the second ATP.
Step 3. A related dehydration/rearrangement step, catalysed by enolase, converts 2-phosphoglycerate to phosphoenolpyruvate at step 9; while this itself is a dehydration rather than a phosphate-transfer step, it is required to set up the final dephosphorylation at step 10. …
